JUNIOR-PAM Chlorophyll Fluorometer for Teaching

Reference
JUNIOR-PAM
Brand
Walz

A miniaturized fluorometer that performs the full range of photosystem II saturation pulse analysis. All optical and electronic components fit in a small 11.5 x 6.5 x 3.0 cm box, and a single 50 cm plastic fiber with 1.5 mm diameter links it to the sample.

One blue power LED (460 nm) handles pulse-modulated fluorescence excitation, actinic illumination and saturating pulses, while a far-red LED provides selective PS I excitation and determination of the Fo’ fluorescence level.

Overview

Leaf clips included

Two leaf clips come with the instrument: a 60° clip for measuring under ambient light and a magnetic clip for Fo-Fm and Fv/Fm measurements.

BLUE version (JUNIOR-PAM optoelectronic unit)

  • A blue LED peaking at 445 nm excites modulated fluorescence; modulation runs from 5 to 25 Hz in 5 Hz steps, or at 100 Hz, and fluorescence is read above 630 nm
  • Actinic light comes from the same LED as the modulated light: at 1 mm from the tip of the 50 cm JUNIOR-PAM light guide it delivers 25 to 1500 μmol m-2 s-1, and saturation pulses reach a maximum PAR of 10,000 μmol m-2 s-1

WHITE version (JUNIOR-PAM/W optoelectronic unit)

  • A white LED with emission peaks at 445 and 545 nm, with the same modulation (5 to 25 Hz in 5 Hz steps, or 100 Hz); fluorescence is read above 650 nm
  • Actinic light from the same LED: between 25 and 1500 μmol m-2 s-1 at 1 mm from the tip of the 50 cm JUNIOR-PAM light guide; saturation pulses up to a maximum PAR of 10,000 µmol m-2 s-1

Shared by both versions

  • Aluminum housing with a USB type B socket, an M8 4-pole socket for the JUNIOR-PAM monitoring leaf clip (accessory), a port for the JUNIOR-PAM light guide and a swivel-mounted sample support
  • Far-red LED peaking at 745 nm for selective excitation of photosystem I
  • PIN photodiode behind a long-pass filter, with a selective window amplifier for pulse-amplitude modulated (PAM) fluorescence
  • Standard USB communication over a 1.2 m type A-B USB cable, which also carries the 5 V DC supply
  • Consumption: 100 mW in normal operation and 500 mW during a saturation pulse
  • Size: 11.5 cm x 6.5 cm x 3 cm (L x W x H); weight: 200 g
  • Works from 0 to + 40 °C, at 35 to 85% RH to avoid condensation

WinControl-3 software

  • WinControl-3 is the system control and data acquisition program (Microsoft Windows XP/Vista/7/8/10) for running the measuring system from a PC and acquiring and analyzing data
  • Saturation pulse analysis. Measured: Ft, F0, FM, F, F0’ (also calculated), FM’; plus PAR, leaf temperature and relative humidity with the JUNIOR-BD monitoring leaf clip. Calculated: F0’ (also measured), FV/FM and Y(II) (maximum and effective photochemical yield of PS II, respectively), qL, qP, qN, NPQ, Y(NPQ), Y(NO) and ETR (electron transport rate)
  • Two fitting routines find the cardinal points α, Ik and ETRmax of light curves
  • Fully programmable with a batch file language
  • Computer: 1 free USB socket, 1 GHz processor, 512 MB RAM, 1024 x 600 pixels screen resolution, USB 2.0/3.0 interface
