The DUAL-PAM-100 relies on a single pair of near-infrared (NIR) wavelengths to follow absorption changes caused by redox changes at the PS I reaction center. The DUAL-KLAS-NIR works with four NIR pairs, which lets it tell apart, without ambiguity, the redox changes of PC, P700 and Fd.
A novel analytical method gives the instrument the in vivo spectral signatures of pure PC, P700 and Fd; that spectral information is what makes online monitoring of their redox changes possible.
Just like the DUAL-PAM-100, the system also measures chlorophyll fluorescence by PAM, with green or blue measuring light. Green light reaches further into the leaf than blue, so green-excited fluorescence carries information from the deeper leaf layers and pairs well with NIR absorption readings, which always cover the whole leaf.
Much of the software is shared with the DUAL-PAM-100, so existing DUAL-PAM-100 users get started with little effort. Automated measuring routines make even complex protocols easy to run.
